In animal studies, large doses of isoflavones have been linked to a higher risk of cancer.

鈥淪omething we hear very often is that people have a lot of concern about consuming soy because there are so many conflicting messages out there,鈥 says Gabrielle Viscardi, a second-year PhD student in the department of nutritional sciences and the study鈥檚 lead author.

This is despite the fact that several health advocacy groups, including the Canadian Cardiovascular Society, already recommend soy foods as part of a healthy diet. Health Canada and the U.S. Food and Drug Administration have both recognized soy 鈥 a high-quality and complete source of protein 鈥 as effective in reducing the risk of heart disease

Soy foods have also been shown to alleviate hot flashes associated with menopause, which affect many women and impact their quality of life.

The trials considered for the 茄子直播 study followed postmenopausal women from around the world who had consumed either soy isoflavones or a non-isoflavone control for at least three months.

The researchers concluded that consumption of soy isoflavones did not affect the four key estrogen-related markers, namely: thickness of uterus lining, vaginal maturation index (a measure of estrogen status) and levels of circulating estrogen and follicle-stimulating hormone.

Their findings support the idea that soy isoflavones behave differently from human estrogen, particularly when it comes to cancers that depend on estrogen to develop. 鈥淲e have estrogen receptors throughout our bodies but, contrary to the hormone estrogen, isoflavones from soy don鈥檛 bind to all the estrogen receptors equally,鈥 says Viscardi, who is also a registered dietitian. 鈥淭hat鈥檚 why we see a beneficial effect on the cardiovascular system and no effect on the female reproductive system.鈥

This difference in biological activity explains why soy isoflavones have been considered as a possible alternative to hormone replacement therapy (HRT), which is used to treat menopause symptoms by replacing the estrogen that the body stops producing during this period.

Chiavaroli notes some people don鈥檛 want to take HRTs and are seeking alternative options.

Further, HRTs may also not be a good option for people at an increased risk of estrogen-sensitive cancers like breast cancer, as well as those with a history of heart disease and stroke. It is also the most challenging to treat due to the resistance of glioblastoma cancer stem cells, from which tumours grow, to therapy. Cancer stem cells that survive after a tumour is treated go on to form new tumours that do not respond to further treatment.

A key finding of the research is that the variability among glioblastoma cancer stem cells can be observed across a gradient between two cell subtypes. On one end is the developmental subtype, which resembles cells in which normal neurodevelopment has gone awry. On the other is the injury-response subtype, which is an inflammatory state. The aim of the study was to identify potential treatment methods to target each subtype, thereby tackling tumours in a more holistic manner.

This study follows earlier research published in Cell Reports that identified vulnerabilities in glioblastoma cancer stem cells that impact their sensitivity to chemotherapy. The next step was to study how vulnerabilities in glioblastoma cancer stem cells vary in a large and diverse set of patient-derived cell lines to identify the most common of these vulnerabilities in each of the subtypes.

The team performed screens in glioblastoma stem cell lines from 30 patients, making this the largest screening study of its kind. The patient-derived cell lines were generated by the lab of Peter Dirks, chief of the division of neurosurgery at SickKids and a 茄子直播 professor of surgery and molecular genetics in the Temerty Faculty of Medicine. Within the cancer stem cell samples, the team found genes responsible for the proliferation of the two cell subtypes that could be targeted to prevent tumour growth. Combining drugs to target both cell subtypes simultaneously could potentially make for a more effective glioblastoma treatment.

鈥淎 lot of the research on glioblastoma is conducted with a limited number of immortalized cell lines grown in serum,鈥 said Fatemeh Molaei, co-first author on the study and graduate student at the Donnelly Centre and the Leslie Dan Faculty of Pharmacy. 鈥淭hese cells aren鈥檛 the best model as they don鈥檛 resemble true glioblastoma cells as much as we would like. The findings from our study represent what we see in a patient鈥檚 tumour more accurately because our cell lines are derived directly from a large group of patients.

鈥淚t鈥檚 through our screens of this group of cell lines that we were able to identify the OLIG2 and MEK genes as drug targets for the developmental cell subtype and the FAK and B1-Integrin genes as targets for the injury-response subtype.鈥

St茅phane Angers, principal investigator on the study and director of the Donnelly Centre, said it had already been established that there are different subtypes of glioblastoma stem cells, but that their differences are not currently being addressed in the clinic.

鈥淚n the future, our results will help in designing new treatments that are tailored to patients by targeting the predominant cell subtype, or both subtypes simultaneously,鈥 said Angers, who is also a professor in the Leslie Dan Faculty of Pharmacy and Temerty Faculty of Medicine. 鈥淭he ability of glioblastoma to adapt to therapeutic treatment is its greatest strength and our biggest challenge. Our study increases our understanding of this type of cancer and proposes a different approach to treating it that will hopefully improve the prognosis of patients.鈥

This research was supported by the Canadian Institutes of Health Research.

News home page banner

Off

Thu, 21 Nov 2024 14:21:00 +0000 Christopher.Sorensen 310649 at 茄子直播 ranked among top 10 universities globally in five subjects: ShanghaiRanking Consultancy /news/u-t-ranked-among-top-10-universities-globally-five-subjects-shanghairanking-consultancy 茄子直播 ranked among top 10 universities globally in five subjects: ShanghaiRanking Consultancy

Christopher.Sorensen Mon, 11/18/2024 - 15:30

Cutline

Mariam Matti

Topic

Global Lens

Temerty Faculty of Medicine

Dalla Lana School of Public Health

Faculty of Arts & Science

Global

Meric Gertler

Rankings

Rotman School of Management

Shanghai Ranking Consultancy

Sociology

The University of Toronto has placed among the top 10 universities globally in five subjects 鈥 sociology (2nd), medical technology (5th), public health (7th), finance (8th) and management (10th) 鈥 in the latest ShanghaiRanking Consultancy rankings by subject.

茄子直播 also ranked in the top 100 globally in 42 subjects in the Global Ranking of Academic Subjects 2024, released last week 鈥 a feat matched only by the University of Michigan.

Among Canadian universities, 茄子直播 ranked first in 27 subjects. Led by post-doctoral researcher Golam Saffi and former master鈥檚 student Lydia To, the team found that INPP4B exerts its tumour-promoting effects via a cellular organ called the lysosome.

鈥淐lassically, the lysosome is a garbage disposal organelle where old and tired proteins and other organelles get degraded to be used for energy and other building blocks for the cell,鈥 says Salmena.

In most cells, lysosomes typically cluster around the nucleus. But in pancreatic cancer cells, the researchers found that INPP4B drove the lysosomes from the cell interior to the periphery, where these organelles fuse with the cell鈥檚 outer membrane. In doing so, the enzymes and other lysosomal factors responsible for breaking down cellular waste are dumped into the space surrounding the tumour cells.

This space contains a network of proteins and molecules that provides crucial structural support to cells and tissues while also restricting a cell鈥檚 ability to move. The release of the lysosome鈥檚 protein-degrading contents into this extracellular space causes the stabilizing network to fall apart, thus making it easier for pancreatic cancer cells to migrate and invade other tissues.

Crucially, Salmena and his team also identified the signalling pathway by which INPP4B drives the movement of lysosomes to the cell edge. INPP4B works with two other proteins 鈥 PIKfyve and TRPML-1 鈥 to modify the lysosome鈥檚 surface structure and alter local calcium levels so that the organelle is propelled to the cell periphery.

Based on these findings, the researchers are testing two experimental drugs that target TRPML-1 and PIKfyve in a preclinical model of pancreatic cancer. They are also studying how the release of lysosomal contents can change the immunological environment of the cancer cells, and what effects that might have on the immune system鈥檚 ability to respond to the tumour.

Salmena first became interested in INPP4B when, during his post-doctoral research, he found that it was involved in breast cancer. Since then, he and his team have shown that the effects of INPP4B vary depending on the context.

For example, in some breast cancer types, INPP4B behaves as a tumour suppressor whereas it has an activating role in other aggressive cancers like pancreatic cancer 鈥 which the Canadian Cancer Society expects to be the third leading cause of cancer death in Canada in 2024, with an estimated 6,100 people dying from the disease.

Salmena and his colleagues later showed that among all cancers, INPP4B levels are highest in pancreatic tumours, and that high levels of the protein are associated with decreased overall survival in people with pancreatic cancer.

The study was a collaboration between Salmena鈥檚 group, Roberto Botelho, a professor of chemistry and biology at Toronto Metropolitan University, and Steven Gallinger, a hepatobiliary and pancreatic surgical oncologist and clinician-scientist at Princess Margaret Cancer Centre, University Health Network and a professor of surgery and laboratory medicine and pathobiology in Temerty Medicine. It also found people who lived with a bird at the time of study were more likely to develop the disease.

Caused by inflammation in the gastrointestinal tract, Crohn鈥檚 disease can have far-reaching consequences on overall health and well-being. Its incidence among children under 10 has doubled since 1995, while the annual cost of inflammatory bowel disease to Canada is estimated at $5.4 billion per year, according to Crohn鈥檚 and Colitis Canada, a national non-profit.

The likelihood of getting Crohn鈥檚 is strongly influenced by genetics, but the environment also plays a role, says Croitoru, who holds the Canada Research Chair in Inflammatory Bowel Diseases. While we can鈥檛 change our genes, we can modify our surroundings and diet, for example, to potentially prevent the disease from occurring.

These findings come from the Genetic, Environmental and Microbial (GEM) Project, the largest study of its kind that seeks to identify potential triggers of Crohn鈥檚 disease.

Coordinated at Mount Sinai Hospital since 2008, the GEM Project has been collecting comprehensive medical and lifestyle data from over 5,000 healthy first-degree relatives of people who have Crohn鈥檚 and come from all over the world, including Australia, Canada, Israel, New Zealand, the U.K. and the U.S.

The project is funded by Crohn鈥檚 and Colitis Canada, the Canadian Institutes of Health Research and the Helmsley Charitable Foundation.

In the 15 years since the study began, over 120 people have developed the disease. 鈥淏y understanding what is different about those who develop the disease, we should be able to predict who is at risk,鈥 says Croitoru.

Previously, the group identified differences in the microbiome and other biomarkers in people who go on to develop Crohn鈥檚 and those who don鈥檛.

In another recent report published in Gastroenterology, Sun-Ho Lee, a clinician-scientist at Mount Sinai Hospital鈥檚 IBD Centre and an assistant professor at the Institute of Medical Science at Temerty Medicine, used available data and machine learning to develop an 鈥integrative risk score鈥 that predicts the risk of Crohn鈥檚 with a high degree of accuracy.

But risk prediction is only the first step, says Croitoru, whose ultimate goal is to be able to intervene and prevent the disease from starting.

He and his team are now conducting research that seeks to devise and test strategies for prevention by, for example, adding supplements to the diet to promote a healthy microbiome.

鈥淪inai Health is committed to groundbreaking research and bringing those discoveries to patients," said Anne-Claude Gingras, director of LTRI and vice-president of research at Sinai Health.

鈥淏y integrating genetic, environmental, and microbial data, Dr. Croitoru and colleagues are paving the way towards personalized intervention strategies that could significantly reduce the incidence of Crohn鈥檚 disease.鈥

News home page banner

Off

Thu, 26 Sep 2024 14:44:39 +0000 rahul.kalvapalle 309604 at AI tool reduces risk of unexpected hospital deaths by 26 per cent: Study /news/ai-tool-reduces-risk-unexpected-hospital-deaths-26-cent-study AI tool reduces risk of unexpected hospital deaths by 26 per cent: Study

Christopher.Sorensen <time datetime="2024-09-23T12:50:13-04:00" title="Monday, September 23, 2024 - If you could predict one thing that AI would tell you, what should that be?鈥 says Verma, who is also the Temerty Professor of AI Research and Education in Medicine.

鈥淎nd one of the leading priorities of everyone was to be able to predict in advance which patients might become so sick in hospital that they need ICU or might die.鈥

During the 19-month-long intervention period, 482 patients in the general internal medicine became high-risk, compared with 1,656 patients who became high risk in the 43-month-long pre-intervention period. There were also fewer non-palliative deaths in the CHARTWatch group than in the pre-intervention group (1.6 per cent versus 2.1 per cent).

鈥淚f I were a patient, I would be so relieved to know that we have this kind of system,鈥 said co-author Yuna Lee, division head and general internist at St. Michael鈥檚 and professor in the department of medicine at 茄子直播.

鈥淪o, when the patient gets a high-risk alert, they鈥檙e going to be assessed by senior staff right away. Also, they鈥檙e going to have quite close monitoring by nursing staff, so they check on them every one to two hours.鈥

Expanding the impact of CHARTWatch

CHARTWatch inputs more than 100 aspects of a patient鈥檚 medical history and current health status that are routinely stored in the hospital鈥檚 electronic medical record. It analyzes the interactions between these inputs and how they change over time. With that information, it鈥檚 able to categorize each patient by their risk for deterioration and send an alert to prioritize treatment.

The study is one of the first to evaluate how CHARTWatch can benefit hospital patients in Canada and shows the potential real-world impact of AI on the health-care sector. Damian Jankowicz, Unity Health鈥檚 vice-president and chief information and AI officer, says he hopes AI tools such as CHARTWatch will continue to have a profound impact on patients.

鈥淗opefully with reduced administrative burden on our providers, they will have more time to spend with our patients and really focus on the patient needs,鈥 he says.

鈥淚 hope that AI will be able to distill the incredible amounts of information that鈥檚 coming at our clinicians into important components and really bring their clinical judgment to the forefront.鈥

This story first was first published by Unity Health.

News home page banner

Off

Mon, 23 Sep 2024 16:50:13 +0000 Christopher.Sorensen 309507 at
